NSW Railways issued tenders for forty 1800hp main line locomotives in 1960 to cover an anticipated increase of traffic on the Main South and to continue the dieselation of the system. A.E. Goodwin won the contract with the Alco 'World' model DL541 - the 45 Class. Following several teething problems, 4501 was in service 31 May 1962, with the last in the class (4540) in the system in February 1964.

In many ways, the 45 Class is a rebodied 44 Class, sharing many components and the engine. The 45 was geared lower than the 44 to fulfill its' intended role as a freight locomotive, however the 45 did see service as a passenger locomotive on many occasions. Like the 44 Class, the 45 Class is one of few main line locomotives to have been seen on almost every line in the state, including Oberon and Dorrigo.
When introduced, the 45 Class served the Main South to Albury and the North Coast to Brisbane. Several years later a number of 45s moved to the west to Bathurst, but it was in the north of the state that the 45 Class made its mark. Eventually all 45 Class locomotives worked out of Broadmeadow on almost every type of service; bulk coal and wheat trains, passenger services, express freight services to Brisbane, the pick-up goods to Tenterfield and banking other trains at Ardglen Bank.

The 45 Class served in regular service until 1994 when, due to faults and the delivery of the 82 and 90 Class locomotives, they were all placed into storage and eventually sold or scrapped. A number were purchased by private operators and have returned to the lines with Silverton, Patrick Portlink and BHP.
